Orthogynest Depot is out of the trade. Which basis to use for a "loco" preparation?

PEG 4000 mixed with PEG 1500/300? What proportion? How to get depot operation? Adding Aerosil 2%? Does a preservative have to be added? May I use a suppository form instead of ovule form because I do not have it and it is the first time in 15 years pharmacists profession that I have to make ovulen. 2 G?

Answer

As suppository base you can use PEG 400 15 g; and PEG 4000 35 g. You also wish to have a depot effect. To do this, the company uses ethoxylated heptanoate, which can be replaced by Cetyl wasesters. I have not been able to find an analogue example and therefore a bit must be gambled for the concentration. I would suggest 5%?   You can also add 2% Aerosil. If we look at nature from the base, a preservative is superfluous! You may, for lack of better, use a suppository form of 2 G. It is quite obvious that the client should be reminded of the abnormal use of these suppositories!!
