How to prepare a mercaptopurine suspension 10mg/ml. ?

Normally one begins with the preparation of the excipient.

For 100 ml excipient the composition reads: Citric acid 75 mg; Bentonite (or better veegum) 1 g; NaCMC   1g; Nipagine 75 mg; Sugar syrup 27 g and freshly boiled water up to 100 ml.  

But I note that Puri-nethol tablets, which you will use as a source of mercaptopurine, contain the following   excipients: lactose, corn starch, hydrolysed starch, stearic acid and magnesium stearate.   These tablets should be be shattered; The easiest maybe with an electric coffee grinder. After grinding seven and some achterbljft on the sieve back grind.

I cannot find any data on the reaction of hydrolysed starch with water? Does gel form??  

So bring in a mortar 75 mg citric acid and 75 mg nipagine and mix with the fine powder, obtained by grinding and sieving of the tablets. Add fresh 27 g sugar syrup into small portions and rub   after each addition. Then add stirring well 40 g freshly boiled water in small quantities and mix after each addition. Check that the whole is viscous by swelling of the hydrolysed starch. Pour the contents of the mortar into a measuring cylinder and apply to volume. Rinse the mortar with the water needed to bring it to volume. Apply in a dark brown red bottle.

If the whole is insufficient viscous so that, after shaking the bottle, the undissolved substance settes too quickly then 1 g of NaCMC should be added. The simplest is to weigh 1 g of NaCMC in a beaker and pour half of the bottle into the beaker. Place in the beaker a blender and mix for a few minutes. Check that no clubbers are in the suspension  !   Add the rest of the bottle under mixes.  

In contrast to the suspension, which has been commercialised in the Netherlands, I have not added any flavourings.

In the composition, about which I have no flavouring. You will have to judge whether this should be? If necessary, we try to correct the taste with a refunded flavour Corrigens: banana aroma,....?
