Introduction

PLO Gel is the abbreviation for Pluronic Lecithin Oil Gel

Pluronic is a brand name for Poloxamer. Poloxamers are non-ionogenic copolymers consisting of a central hydrophobic chain of polypropylene oxide with a hydrophilic chain of polyoxyethyleenoxide on both sides.

The ratio between the propylene oxide part and the polyoxyethyleeoxide part determines the HLB value. These poloxamers find frequent applications as humidifiers (lens products), emulsifiers, Solubilizers and also as gelformers.

The PLO Gel uses Pluronic F127   or Poloxamer 407,   consisting of a   Polyoxyethyleenketen   with 101 ethylene oxide monomers and a polypropylene oxide chain consisting of 56 polypropylene oxide units.

This gel as well as the constituents needed for the preparation of the PLO gel were taken from the market a dozen years ago.

The main application of this gel is for transdermal administration of drugs both in humans and animals (mainly cats). There is a certain contradiction regarding the value of this gel as excipient for transdermal administration. Nevertheless, certain scientific articles have shown that the active ingredient is effectively absorbed and enters the bloodstream. Gabapentin, ketamine, testosterone and other corticosteroids are examples of active ingredients that can be administered through this pathway.

To offer the pharmacists the possibility to deliver a preparation when the physician prescribes a composition, which he usually takes over from a study in the US published, an alternative formula was developed. The name CLO Gel was given to this composition.

THE CLO Gel

In The CLO gel is also used lecithin. Lecithin is also no longer available in Belgium as a pharmaceutical raw material. But the dietary supplement Mannavital contains soy lecithin.  

The preparation consists of three steps

1 ° Step: Preparation solution lecithin in liquid wax

  • Rub 12 g of Mannavital grains finely into a mortar; The wall is well-off because lecithin is sticky. Weigh 10 g of lecithin powder and mix it with 10 g isopropyl. Mix to dissolve  , possibly under light heating; Then stay overnight.

2 ° Step: Preparation of Carbomer gel

  • Carbomeer 1 G, tromethamol 1 g, sorbic acid 250 mg and water up to 80 g

3 ° Step

  • dissolve the medicine in the Lecithin solution   and mix this solution with the Carbomeergel. Treatment with the Unguator enhances the texture and color of the prepared CLO gel.
